I have a library of ~40,000 images, including 1/3 digital images I've shot and made smart previews on import, and 2/3 scanned and from other sources, most of which do not have smart previews. Is there any reason I can't select all of them and create smart previews, or will that screw up something about those that already have them?

No, that won't screw up anything.

One more -- if I do Smart Previews, there is pretty much no reason to also do 1:1, is there?
Smart Previews and 1:1 previews are completely unrelated.

Smart Previews are used only in the Develop module, and they allow the user to edit images when the originals are offline. They can also be used instead of originals for performance reasons even when the originals are online.

1:1 previews are used in the Library module, where they allow the image to be zoomed to 1:1 without delay (if only standard previews exist, there would typically be a loading delay when zooming to 1:1, as the 1:1 preview has to be created first). Note that when an image is edited, the existing library previews are erased (because the edits make them obsolete), and only a standard preview is automatically rebuilt when the image is next accessed in the Library module.
